I’m at a loss at what to do. I’ve restarted my computer, uninstall and reinstalled Synth V. The program will not run any previous files or new ones I created. When I hit the playback button, the playhead doesn’t move.

When I run my DAW (Bitwig) and then try to run SynthV in stand-alone mode SynthV will do something similar - playhead frozen with a RED play icon.
It took a while to figure out that the DAW was taking exclusive control of the audio system.
Bear with me on this…
In SynthV open the Settings tab and look at audio settings, maybe SynthV is trying to use the wrong audio system or output device.
My only suggestion is to revert to Sonoma from a backup. I am using Sonoma with Logic with no issues (except that ARA is not functional, which is a dreamtronics programming flaw that can be worked around).
I don’t mean to sound supercilious, but you should NEVER upgrade your Mac OS if the current one works. Sequoia is only a few months old and is still quite buggy. A good practice is to only upgrade your OS when the current one no longer runs the software you want to run. I only upgraded to Sonoma from Monterey a few months ago. I’ve learned this the hard way, like you just did
Never upgrade to the latest OS until it has been out at least one year. Let other people be the beta testers!
Another good practice is to check with the software manufacturer if what you use is going to be compatible, AND check on user forums for real users’ experience.
